Has anyone else spent ages having an absolute c*** of a time trying to get the drive back in?
Im sure Im at TDC on #1, when I am I cant for the life of me get it to thread.

When I rotate the crank out of the way slightly it slips in fine but I'm no longer on TDC

You don't need to put the drive in on TDC. Put the drive in where the crank is convenient, and then roll the crank back to TDC to check your position. If it is not right, repeat the process, but rotate the drive before putting it back in so it lines up better next time you put it back to TDC. Just do that a couple of times and you will get the setting you are after.

Its amazing what coming back to a problem later will solve.
Worked it out rotating the engine back ad forth for a while.

Right so this is really doing my head in.
Distributor shaft is set to 1 o'clock at TDC, distributor is in with rotor pointing to 1, I have spark have fuel but still can't get the bugger to run.
I'm getting big backfires out the carb.
I have tried advancing the timing, I have tried the mixture screw on the carb.

Could the timing be 180 out? Surely not. After all this.
Is it possible I was at TDC on the wrong stroke?

Sounds like you are 360degrees of the crank out (180 at the dizzy)

When you are using the manual to build your engine, you time the camshaft in with no.4 cylinder firing. The image showing the positioning of the distributor drive is for no.1 cylinder firing.

You can either put the shaft in the correct spot, or just change your spark plug leads around.
